Background: Phosphodiesterases (PDEs) regulate the intracellular levels of cAMP and cGMP by hydrolyzing cAMP and cGMP to their inactive 5' monophosphates. These cyclic nucleotides play an important role as second messengers in diverse physiological functions. PDE10A plays a key role in signal transduction by regulating the intracellular concentration of cyclic nucleotides. PDE10A hydrolyzes both cAMP and cGMP to nucleoside 5?-monophosphate, but exhibits higher affinity for cAMP, and it utilizes cAMP more efficiently as a substrate (Fujishige K. et al., 1999).

Description: Recombinant HEK293 cell line expressing human PDE10A2 (phosphodiesterase 10A, accession number NM_006661).
N-terminal His-tagged human PDE10A2 has been stably expressed in a human embryonic kidney (HEK293) cell line. PDE10A2 expression was confirmed by Western blotting. The function of PDE10A2 was confirmed by cAMP detection assay. Forskolin is commonly used to raise levels of intracellular cyclic AMP (cAMP). When HEK293 cells are activated by forskolin, cAMP levels are upregulated in parental cells, whereas cells overexpressing PDE10A show reduction in cAMP accumulation. Inhibition of PDE10A activity restores cAMP levels.
