Background: SARM1 is the executioner of the axonal degeneration pathway that promotes depletion of axonal NAD+. SARM1 functions as an ADP-ribosyl cyclase and nicotinamide adenine dinucleotide (NAD) glycohydrolase. Dimerization of SARM1-TIR domains in neurons induces axonal degeneration since SARM1-TIR domains have intrinsic NADase activity which cleave NAD+ into ADP Ribose (ADPR), cyclic ADPR, and Nicotinamide. Depletion of axonal NAD+ triggers pathological axon loss. Therefore, inhibition of SARM1 NAD+ cleavage activity may potentially reduce axonal degeneration.
