Background: Rat small intestinal smooth muscle cells were isolated from small intestine tissue. The small intestine is located in the abdomen. The upper end is connected to the pylorus and the stomach, and the lower end is connected to the large intestine through the appendix. It is the main place for food digestion and absorption. The small intestine is coiled in the abdominal cavity, connected to the stomach pylorus at the upper part, and the cecum at the lower part. It is divided into three parts: duodenum, jejunum and ileum. Digestion in the small intestine is very important, because after the chemical digestion of pancreatic juice, bile and small intestinal juice in the small intestine and the mechanical digestion of small intestine movement, the digestion process is basically completed, and nutrients are absorbed by the small intestinal mucosa. The wall of the small intestine is composed of mucosa, submucosa, muscle layer and serosa. Its structure is characterized by ring-shaped folds on the tube wall and many villi on the mucosa. The epithelium at the root of the villi sinks down to the lamina propria to form a tubular intestinal gland with the opening between the roots of the villi. Villi and intestinal glands are closely related to the digestive and absorption functions of the small intestine. The cells that make up the intestinal glands include columnar cells, goblet cells, Paneth cells and undifferentiated cells. Columnar cells and endocrine cells are similar to villi epithelium. Columnar cells close to villi are similar to absorber cells. Columnar cells in the deep part of villi have few and short microvilli and do not form striated borders. The small intestine has three functions, namely digestion, absorption and secretion, and motor functions, of which absorption and secretion are the main functions. The contraction of smooth muscle cells is responsible for the power of intestinal peristalsis, which promotes the downward movement of food. After 3 days of primary isolation and culture of small intestinal smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network. When the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. Leiomyosarcoma of the small intestine is a malignant tumor originating from the muscular layer of the small intestine wall, the submucosal muscular layer and the vascular smooth muscle of the intestinal wall. It is the most common type of malignant tumor of the connective tissue of the small intestine. Therefore, the culture of small intestinal smooth muscle cells in vitro provides the basis and premise for the study of small intestinal leiomyosarcoma. In addition, because the influencing factors are relatively single, in vitro cell culture is the basis for studying cell function and the corresponding cell signal transduction mechanism. The rat small intestinal smooth muscle cells produced by our company are prepared by the trypsin-collagenase combined digestion method and the differential adhesion method. The total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA immunofluorescence identification, the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
