Background: Rat muscle-derived stem cells are isolated from muscle tissue. Muscle-derived stem cells (MDSCs) are adult pluripotent stem cells with multi-cell lineage differentiation and self-renewal capabilities. By inducing stimulation, MDSCs can differentiate into a variety of cell and tissue types such as fat, bone, cartilage, smooth muscle and nerve cells. As a donor cell for gene therapy and tissue engineering, it has become a research hotspot in the treatment of myocardial infarction, Duchenne's muscular dystrophy and other diseases. In recent years, the application of muscle-derived stem cells in the treatment of urinary incontinence has gradually gained attention. It should be noted that muscle-derived stem cells mainly exist in skeletal muscle tissue, and are only a few cells of mature tissue. Usually in a static state, it will only activate when cell stress and tissue loss. And as the age increases, the number of cells contained gradually decreases, and the current treatment of clinical diseases is mainly for adults. The in vitro expansion of muscle-derived stem cells is essential for cell transplantation and stem cell-mediated gene therapy. Studies on the expansion of myogenic stem cells have found that EGF, FGF-2 and SCF can stimulate cell proliferation through meiosis or promote non-dividing cells into the mitotic cycle. More importantly, cytokine-induced in vitro expansion can maintain stem cell phenotype by self-renewal without stimulating cell differentiation. The rat muscle-derived stem cells produced by our company are prepared by the combination of collagenase-neutral protease-trypsin digestion method, density gradient centrifugation, differential adhesion method, and finally through the culture and selection of the special medium for muscle-derived stem cells. The total amount is about 5×10^5 cells/vial. The cells are identified by CD34, Desmin, and CD45 immunofluorescence, and the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
