Background: Rat esophageal smooth muscle cells are isolated from esophageal tissue. The esophagus can be divided into cervical, thoracic and abdominal segments. The cervical segment of the vertebrate esophagus is located behind the trachea and the front of the spine, and the thoracic segment is located in the mediastinum between the left and right lungs. The thoracic segment is connected to the abdomen in the abdominal cavity through the diaphragm foramen, and the abdominal segment is very short to connect to the stomach. The mammalian esophagus is divided into four layers from the inside to the outside: mucosal layer, submucosa, muscle layer and adventitia. Among them, the upper 1/3 of the muscular layer is composed of skeletal muscle, the lower 1/3 is smooth muscle, and the middle section is a mixture of skeletal muscle and smooth muscle. Among them, the arrangement of muscle fibers is divided into two layers: inner ring and outer longitudinal. The peristalsis of the esophagus is strictly controlled by the contraction of the esophageal smooth muscle. There is also a sphincter in the esophagus. The upper esophageal sphincter is located at the level of the cricoid cartilage. Located at the lower end of the esophagus, one part is on the diaphragm, passing through the diaphragm foramen, and the other part is under the diaphragm, which is called the lower esophageal sphincter. After 3 days of primary isolation and culture of esophageal smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network; when the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. The rat esophageal smooth muscle cells produced by our company are prepared by the trypsin-collagenase combined digestion method and the differential adhesion method. The total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA immunofluorescence identification, the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
