Background: The rat colonic smooth muscle cells were isolated from the colon tissue. The colon continued to the cecum in the right iliac fossa and connected to the rectum at the 3rd sacral plane. The colon is divided into 4 parts: ascending colon, transverse colon, descending colon and sigmoid colon, most of which are fixed on the posterior wall of the abdomen. The arrangement of the colon resembles the English letter "M" and surrounds the small intestine. From the inside to the outside, the cross section of the colon is: mucosa (epithelial layer, lamina propria, muscularis mucosa), submucosa (loose connective tissue), muscularis (two smooth muscles in the inner ring and outer longitudinal), and adventitia (fibrous membrane) or serosa). Colonic smooth muscle cells are mainly distributed in the inner circular and outer longitudinal smooth muscles of the muscularis mucosa and the muscularis. The movement of the colon is small and slow, and the response to stimulation is also slow. Colonic smooth muscle plays an important role in the research of food digestion and absorption, intestinal movement and substance secretion, inducing systemic inflammation, and intracellular signal transduction pathways. After 3 days of primary isolation and culture of colonic smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network; when the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. The motor activity of colonic smooth muscle is regulated by neurotransmitters, gastrointestinal hormones and drugs. With the development of modern gastrointestinal kinetics research, the research on gastrointestinal motility has developed from the organ and tissue level to the cell and molecular level. It is required to complete various electrophysiology, pharmacology and molecular biology on a single smooth muscle cell specimen of the gastrointestinal tract. In addition, because the influencing factors are relatively single, in vitro cell culture is the basis for studying cell function and the corresponding cell signal transduction mechanism. The rat colonic smooth muscle cells produced by our company are prepared by trypsin-collagenase combined digestion method combined with differential adhesion method. The total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA immunofluorescence. The purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
