Background: Rat brain microvascular endothelial cells were isolated from brain tissue. Cerebral microvascular endothelial cells are the main component of the blood-brain barrier. They are a single layer of flat epithelial-like cells that compose the lumen of the cerebral microvessels, which can restrict soluble substances and cells from entering the brain from the blood. Brain microvascular endothelial cells have some of the same characteristics as peripheral endothelial cells. The biologically active substances it produces and secrete play an important role in maintaining vascular tension, regulating blood pressure, and anti-thrombosis, and have important pathophysiological significance in the pathogenesis of cerebrovascular diseases. Like peripheral endothelial cells, the surface of brain microvascular endothelial cells express cell adhesion molecules, which regulate the entry of white blood cells into the brain. Due to the organ specificity of microvascular endothelial cells, endothelial cells are usually derived from relevant tissues in disease research. Its main characteristics are as follows: 1. There are many tight junctions between the brain microvascular endothelial cells, which produce high transendothelial impedance and delay the flux around the cells. 2. Brain microvascular endothelial cells lack the fenestration structure of endothelial cells, and their liquid phase substance pinocytosis level is low. 3. Cerebral microvascular endothelial cells have an asymmetric localization enzyme and carrier-mediated transport system, resulting in a "polarized" phenotype. The rat brain microvascular endothelial cells produced by our company are prepared by a collagenase-neutral protease mixed digestion method combined with a density gradient centrifugation method, and finally through a special culture medium for endothelial cells.
