Background: Rat bone marrow mesenchymal stem cells (BMSC) are isolated from bone marrow. Bone marrow is the body's hematopoietic tissue, located in many bones in the body. There are two types of bone marrow in adult animals: red bone marrow and yellow bone marrow. Red bone marrow can produce red blood cells, platelets and various white blood cells. Platelets have a hemostatic effect, and white blood cells can kill and inhibit various pathogens, including bacteria and viruses. Certain lymphocytes can make antibodies. Therefore, bone marrow is not only a hematopoietic organ, it is also an important immune organ. Bone marrow is a spongy tissue that exists between the marrow cavity of long bones (such as humerus and femur) and the sparse cancellous bone of flat bones (such as iliac bone). It is a spongy tissue. The bone marrow that produces blood cells is slightly red, called Red bone marrow. At birth, red bone marrow fills the bone marrow cavity of the whole body. With age, fat cells increase. A considerable part of the red bone marrow is replaced by yellow bone marrow. In the end, almost only the flat bone marrow cavity contains red marrow. Bone marrow mesenchymal stem cells in the bone marrow stromal system are stem cells with high self-renewal ability and multi-differentiation potential in addition to hematopoietic stem cells. It can differentiate into bone, cartilage, muscle tissue, skin, fat, nerve and other tissues, so it can be used as seed cells in tissue engineering. In bone marrow, BMSC accounts for 0.001%-0.1% of the total number of bone marrow nucleated cells, and the content is extremely low. Bone marrow mesenchymal stem cells require high in vitro culture conditions. During the culture process, they are affected by such conditions as attachment time, planting density, serum content, culture temperature and medium pH. The rat bone marrow mesenchymal stem cells produced by our company are prepared by washing bone marrow, density gradient centrifugation, and differential adhesion method combined with medium selection. The total number of cells is about 5×10^5 cells/vial. The cells are identified by CD90/CD44 and CD34/CD45 immunofluorescence, and the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
