Background: Rat aortic smooth muscle cells separate the aortic tissue. The aorta is the main artery of the systemic circulation. The path of travel is as follows: the ascending aorta starts from the left ventricle, moves to the height of the second thoracic joint on the right side as the aortic arch, and the arch moves to the left and back to the lower edge of the fourth thoracic vertebral body as the descending aorta. The aortic hiatus that penetrates the diaphragm at the height of the 12th thoracic vertebral body shifts into the abdominal aorta, the upper part is the thoracic aorta, and the lower edge of the 4th lumbar vertebral body is divided into the left and right common iliac arteries. The common iliac artery is divided into internal and external iliac arteries at the height of the sacroiliac joint. After 3 days of primary isolation and culture of aortic smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network. When the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. Aortic smooth muscle cells play an important role in the occurrence and development of cardiovascular diseases. With aortic smooth muscle cells as the experimental research object, exploring the pathogenesis of cardiovascular diseases is the current research hotspot. Aortic smooth muscle cells cultured in vitro are useful for studying their physiological functions, the effects of drugs and pathophysiological changes under the action of various pathogenic factors are of great significance. The rat aortic smooth muscle cells produced by our company are prepared by the trypsin-collagenase combined digestion method and the differential adhesion method. The total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA Immunofluorescence identification, the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
