Background: Rabbit tracheal smooth muscle cells are isolated from tracheal tissue. Trachea, a part of the respiratory organ, a cylindrical tube with a slightly flat back wall. The upper end is flat and the lower edge of the sixth cervical vertebra is connected to the cricoid cartilage. Down to the junction of the fourth and fifth thoracic vertebrae (equivalent to the plane of the sternum angle) is divided into the left and right main bronchus. The bifurcation is called the tracheal bifurcation. The trachea is mainly composed of 14-16 semi-circular cartilage, elastic, and the cartilage is C"-shaped cartilage ring, The gap is backward, the cartilage rings are connected by ligaments, the gap behind the ring is connected by smooth muscle and dense connective tissue, maintaining a continuous open state. The lumen is lined with mucosa, and the surface is covered with ciliated epithelium.The mucus secreted by the mucous membrane can adhere to dust particles in the inhaled air, and the cilia continuously swing to the pharynx to expel the mucus and dust to purify the inhaled gas. Tracheal smooth muscle is one of the important structural components of the trachea and plays an important role in the normal physiological process of the body. It can maintain the airway tension and maintain the tubular shape of the airway, which is conducive to air circulation and maintain lung ventilation. After 3 days of primary isolation and culture of tracheal smooth muscle cells, the cells can be seen to adhere to the flask, and the cell shapes vary in size and are spindle-shaped and irregular.Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network. When the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. The abnormality of tracheal smooth muscle cells is one of the important pathological features of respiratory diseases, and its proliferation and hypertrophy are the key to airway remodeling. The rabbit tracheal smooth muscle cells produced by our company are prepared by the trypsin-collagenase combined digestion method and the tissue patch method, the total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA immunofluorescence, and the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria , yeast and fungi, etc.
