Background: Mouse pancreatic stellate cells are isolated from pancreatic tissue. The pancreas is divided into exocrine glands and endocrine glands. The exocrine glands are composed of acinar and ducts. The acinar secretes pancreatic juice, and the duct is the channel through which pancreatic juice is discharged. Pancreatic juice contains sodium bicarbonate, trypsinogen, lipase, amylase, etc. Pancreatic juice is discharged into the duodenum through the pancreatic duct and has the function of digesting protein, fat and sugar. The endocrine glands are composed of cell clusters of different sizes, namely pancreatic islets. Pancreatic islets are mainly composed of four types of cells: α cells, β cells, γ cells and PP cells. α cells secrete glucagon to raise blood sugar; β cells secrete insulin to lower blood sugar; γ cells secrete somatostatin to inhibit the secretion of α and β cells in a paracrine manner; PP cells secrete pancreatic polypeptide to inhibit gastrointestinal motility, pancreatic juice secretion and gallbladder contraction. Fibrosis is a typical pathological feature of chronic pancreatitis. Activated pancreatic stellate cells (PSC) are the main effector cells of pancreatic fibrosis. The isolation and successful culture of PSC is an important prerequisite for studying pancreatic fibrosis in vitro. The cytoplasm of inactivated PSC is rich in vitamin A lipid droplets and expresses Desmin protein, while the activated PSC expresses alpha-smooth muscle actin (alpha-SMA). PSC has two kinds of resting state and active state, and each has specific markers. The vitamin A lipid droplets rich in the cytoplasm of PSCs in the resting state can be stained red by oil red O and positively express Desmin. The activated PSC positively expresses alpha-SMA. Oil red O staining results showed that after the primary cells were cultured for 6 days, there were still obvious lipid droplets in the cytoplasm. After passage, the orange-red lipid droplet particles were significantly reduced or even disappeared. Immunocytochemical staining results showed that the cells still expressed Desmin 24h after inoculation, and Desmin was almost no longer expressed after 48h. After 48h of culture, most cells began to express alpha-SMA. As the time of culture and the number of passages increased, the cells expressed alpha-SMA instead of Desmin, indicating cell activation. It suggested that the activation process was initiated 24 hours after PSC inoculation, and most of the cells were activated on the 6th day of culture, and the cells were in a highly activated state after passage. Primary pancreatic stellate cells can be used as a cell screening model for new drugs for chronic pancreatitis. Current studies have found that when the pancreas is damaged, the pancreatic stellate cells are activated under the action of various stimulating factors, resulting in changes in cell morphology and function, and promoting the hyperplasia of matrix, massive production of collagen and irregular deposition. The mouse pancreatic stellate cells produced by our company are prepared with collagenase, and the total amount of cells is about 5×10^5 cells/vial. The cells are identified by Desmin and alpha-SMA immunofluorescence, and the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
