Background: Mouse bladder smooth muscle cells were isolated from bladder tissue. The bladder is a hollow organ mainly composed of smooth muscle cells. The relaxation and contraction of bladder smooth muscle allows the bladder to store and discharge urine respectively. The regulation of bladder smooth muscle cell phenotype and the inducible expression of nitric oxide synthase are related to a variety of pathological conditions, including bladder dysfunction. Studies have shown that tissue hypoxia inhibits the proliferation of bladder smooth muscle cells, and the differentiation of bladder smooth muscle cells depends on the factors released by bladder epithelial cells. The secretory phenotype of bladder smooth muscle extracellular matrix can be changed by the frequency of mechanical deformation experienced by the cells. Smooth muscle is a common pathway in many diseases. Therefore, understanding how smooth muscle changes during the occurrence and duration of the disease is an important step in the progress of treatment methods. The bladder wall is composed of three layers of tissue, from the inside to the outside are the mucosal layer, the muscle layer and the adventitia. Among them, the muscle layer is mainly composed of smooth muscle. After 3 days of primary isolation and culture of bladder smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus, cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network. When the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics. Culturing bladder smooth muscle cells in vitro not only provides the necessary means for tissue engineering bladder and urethra to grow cells, but is also the basis and prerequisite for studying leiomyomas. The mouse bladder smooth muscle cells produced by our company are prepared by the trypsin-collagenase combined digestion method and the differential adhesion method. The total number of cells is about 5×10^5 cells/vial. The cells are identified by alpha-SMA Immunofluorescence identification, the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
