Adeno-associated virus (AAV) has emerged as a leading platform for gene delivery for treating various diseases due to its excellent safety profile and efficient transduction to various target tissues. Recombinant AAV (rAAV) vectors are currently being considered for various non-traditional uses such as gene knockdown, genome editing or modification, and non-coding RNA modulation. AAV are non-enveloped, small, single-stranded DNA viruses, capable of infecting both dividing and non-dividing cells. Their genome is about 5 kb and is naturally non-pathogenic to humans. To date, 12 primary AAV serotypes and more than 100 different variants have been identified/discovered. Each serotype has a unique cellular tropism with variations in organ biodistribution as well as transduction properties, possibly due to differences in receptor binding.
